the 2022 meta-analysis in the international journal of molecular sciences pooled the dog and cat trials and placed glucosamine and chondroitin at the bottom, with an effect size (d=-1.39) that actually favoured the placebo groups. of nine trials it examined, only one came out positive - and the reviewers weren't much impressed by it: the improvement rested on a non-validated subjective scoring tool measured at a single timepoint, with the dose cut by a third partway through. the authors' plain conclusion was that these products should no longer be recommended for the stiff, ageing joints they're sold for.
there's a plumbing problem underneath all this. when researchers dosed beagles with glucosamine, oral bioavailability came out at roughly 12% (chondroitin fared worse, around 5%). most of the mouthful never reaches the bloodstream, which makes it hard for even a generous label figure to do much. that doesn't prove glucosamine is useless - plenty of owners are convinced by it, and it's very safe - but it does explain why the controlled evidence is so wobbly.

here's the part that makes glm more than a trend. it isn't a lone isolated molecule - it's whole shellfish, and its make-up does a lot of the work.
